How do you ascertain your new-to-file buyers so that you can market directly to them? I've worked with some reports and T-Stats, but haven't had any luck determining recent purchasers with newly-created accounts.

Try adding in the criteria "constituent create date" when creating your segment.
Constituent create date can work as Joseph pointed out. But this could miss prospects you've added to your database or who signed up to email marketing some time ago. They have never booked but are not recently created constituents.
I usually do an exclusion segment in my extraction has perf date less than or = to the start date I want to begin looking for new buyers. Then any inclusion segment you build below that one in the extraction will be only first time bookings.
I would create a cube in T-STATS of all my ticket buyers with specific single ticket price types. You could even break this down by MOS if you wanted to market differently to say your web buyers versus your phone buyers (if you use different MOS for your different purchase paths). You can then export that as a list into Tessitura. Once there, you can manipulate that data in an extraction however you want. To cover the prospects who haven't purchased anything but want to receive information about shows, you could make an interest that you check so that they get pulled in as well to be marketed to for shows.
